PNAS:新化合物能抗击耐药菌

  诺贝尔奖得主、该研究高级研究作者、化学教授Sidney Altman说,“在当前药物中使用的浓度下该化合物是有效的,并有望用于抗击感染性疾病及耐药性生物。”1989年,Altman和Thomas R. Cech一道发现RNA的催化性能,并因此荣获了诺贝尔化学奖。

  新方法使用一种RNA的片断来抑制造成发病机制的基因表达,该RNA可”编码”以上基因。最终产物在暴露于细胞内的一种酶后,关闭了使细菌存活的基因表达。

  通过致力于细胞培养工作,耶鲁大学科学家发现他们可以使用1种新化合物促进这种RNA在多种细菌细胞中传递,这种新化合物以人类T细胞中的一种肽为基础。它不仅使菌株对当前药物更敏感,而且还能通过靶向于关乎细胞生存的基因杀灭细菌。科学家估计,此方法“靶定”细菌的有效性较现有化合物高出 10~100倍。

  “我们认为该化合物将在老鼠中有效并可以从老鼠转至人类,”Altman说。
