通过流式细胞仪分析血小板糖蛋白(GP)的表达被用于诊断相关的GP相关的血小板减少症。尽管富含血小板的血浆可用于目标事件的明确识别,但这种策略对小样本量或血小板计数低和/或血小板过多的患者不可行。但是,由于难以将血小板与红细胞(RBC)区分开来,因此妨碍了全血(WB)的使用。为了避免这些限制,我们评估了RBC门控策略的可行性。

除了血小板GPIb、GPIIa/IIIa和p -选择素(CD62P),柠檬酸全血(CWB)标本还对红细胞特异性糖素A (CD235a)进行了染色。cd235a阴性的血小板事件通过正向/侧散点特征进一步鉴别,血小板GP表达分析相对于平行处理的健康对照样本。

建立的参考区间可以明确分别在确诊为Glanzmann血栓性贫血或Bernard-Soulier综合征患者样本中GPIIb/IIIa-或GPIb表达模式降低。可以表明,对2500个血小板事件的分析足以进行可靠的GP表达分析,使所提出的方法适用于低血小板计数的样本。

这项研究证明了基于cd235的RBC排除用于CWB中血小板GP表达分析的可行性。与直接染色血小板特异性抗原进行目标识别相比,这种间接门控的方法通常适用于任何潜在的血小板GP表达不足。
