Nature:清华大学丁胜教授利用小分子实现免疫细胞命运调控

来自Gladstone研究所和清华大学的丁胜教授团队在自然杂志发文,他们发现一个名为(氨基氧基)乙酸的小分子,能够将T辅助17(TH17)细胞分化为诱导的调节性T(iTreg)细胞。

现有的研究已经证明,代谢能够和表观遗传,以及转录共同作用,调节细胞的命运以及功能。代谢的改变能够满足T细胞分化的能量以及生物合成需求,但是,至今为止,代谢是否可以通过表观遗传学机制来控制T细胞的命运还是未知的。来自Gladstone研究所和清华大学的丁胜教授团队在自然杂志发文,他们发现一个名为(氨基氧基)乙酸的小分子,能够将T辅助17(TH17)细胞分化为诱导的调节性T(iTreg)细胞。

研究人员发现,转氨作用的增加会使得TH17细胞中2-羟基戊二酸的水平升高。转氨作用主要由GOT1催化。2-羟基戊二酸的积累会导致Foxp3基因位点的超甲基化,并抑制Foxp3转录,这对于TH17细胞的命运决定起着至关重要的作用。如果抑制谷氨酸转化为α-酮戊二酸,可以防止2-羟基戊二酸的产生,进而减少Foxp3基因的甲基化,使得Foxp3表达量增加。这会进一步通过拮抗转录因子RORγt的功能,来阻断TH17细胞的分化,并促进了其向iTreg细胞转化。

(氨氧基)乙酸对GOT1有选择性抑制作用。其能够通过调节TH17和iTreg细胞之间的平衡,改善小鼠模型中的自身免疫性脑脊髓炎的病况。

靶向谷氨酸依赖性代谢途径可能代表了一个针对TH17介导的自身免疫疾病的治疗新策略。
