一种葡萄球菌菌株产生的分子会干扰DNA的合成

来自加州大学圣地亚哥分校的研究人员发现一种在人体皮肤上发现的表皮葡萄球菌含有具有抗癌能力的化学物质。

这一研究成果公布在2月28日的Science Advances杂志上。

文章作者,加州大学圣地亚哥分校Richard Gallo说:“这种菌株的存在可能是为了提供自然保护,未来也许可以被用于治疗,抑制各种形式的癌症生长。”

Gallo和他的同事本来是在研究皮肤细菌的抗菌特性,但他们发现了一种能产生6-N-羟基氨基嘌呤(6-HAP)的表皮葡萄球菌菌株,6-HAP类似于腺嘌呤,也就是DNA的关键成分。

“由于其结构特殊,我们就想知道它是否会干扰DNA合成,”Gallo说。

当研究小组进一步调查时,他们发现6-HAP会阻断DNA聚合酶的活性,DNA聚合酶是合成DNA所需的酶。他们还发现6-HAP能够抑制各种类型的肿瘤细胞如黑素瘤和淋巴瘤的增殖。而且这种物质不会抑制正常人类皮肤细胞的生长,因为正常细胞具有癌细胞上不存在的6-HAP失活酶。

之后研究人员将这种产生6-HAP的细菌用于小鼠皮肤,结果发现用紫外线照射后,小鼠发生恶化前皮肤肿瘤的比率比接受不产生该化学物质的微生物菌株的小鼠更少。

威斯康星大学麦迪逊分校的生物化学家Lindsay Kalan认为,这一发现强调了“微生物组对人类疾病影响的潜力”。不过,她也补充说,“在我们开始进行疾病治疗之前,了解微生物群与其宿主如何相互作用是非常重要的。”
