Blood:衰老诱导的IL27Ra信号可损伤造血干细胞

造血干细胞(HSC)衰老与骨髓增生相关疾病及免疫衰老的风险增加息息相关。

造血干细胞(HSC)衰老与骨髓增生相关疾病及免疫衰老的风险增加息息相关。

在该研究中,研究人员发现衰老相关的炎症反应能够通过TNFα/ERK/ETS1/IL27Ra信号通路促进HSC的衰老过程。

在衰老过程中,TNFα这一众所周知的炎症相关生物标志物的表达水平升高,并通过ERK/ETS1信号通路诱导HSC中IL27Ra的表达。敲除IL27Ra的表达能够挽救HSC的功能衰退及骨髓分化偏向,并逆转TNFα对HSC的抑制作用。

IL27Ra敲除的年老小鼠的髓系分化偏向性HSC比例降低,而在野生型小鼠中该比例没有变化。与IL27Ra-HSC相比,IL27Ra+HSC具有受损重建能力及骨髓分化偏向性,且在炎症损伤后其可以作为髓质恢复库。此外,炎症相关基因在IL27Ra+HSC中富集,且其表达水平随着年龄的增长而升高。

以上研究表明,衰老诱导性IL27Ra信号通路会对造血干细胞造成损害作用;在对抗衰老过程对造血功能的有害影响时,增加对IL27Ra通路的干扰提供了潜在的治疗机会。
