J Periodontol:比较不同组织来源的诱导多能干细胞的牙周分化倾向

诱导多能干细胞(iPSCs)几乎被认为就是胚胎干细胞,但是它们具有残余的体细胞记忆,这使得它们更会表现出供体组织的分化倾向。为了进一步验证这种假设,作者第一次比较了人牙龈成纤维细胞来源的iPSCs (G-iPSCs)和人新生儿皮肤来源的iPSCs (S-iPSCs)的牙周分化趋向,以此评估是否G-iPSCs能够更有效的诱导形成牙周细胞。

诱导多能干细胞(iPSCs)几乎被认为就是胚胎干细胞,但是它们具有残余的体细胞记忆,这使得它们更会表现出供体组织的分化倾向。为了进一步验证这种假设,作者第一次比较了人牙龈成纤维细胞来源的iPSCs (G-iPSCs)和人新生儿皮肤来源的iPSCs (S-iPSCs)的牙周分化趋向,以此评估是否G-iPSCs能够更有效的诱导形成牙周细胞。研究分别使用生长/分化因子-5和结缔组织生长因子诱导G-和S-iPSCs 14天。采用免疫荧光染色和实时聚合酶链式反应检测比较相关标志物的表达水平。并且,在水凝胶载体内培养这些前体细胞,并在不肥胖的严重糖尿病合并免疫缺陷小鼠皮下进行埋植试验。随后,再通过HE、免疫组化染色和预标记PKH67的免疫荧光共区域化进一步观察他们不同的牙周表型。结果显示,正如所预料的,基于体外高表达相应标志物的基因和蛋白水平,2种iPSCs均倾向于分化回其来源组织细胞。HE染色显示,G-iPSCs水凝胶培养后的组成比S-iPSCs水凝胶产生更多的矿化结构。免疫组织化学染色和免疫荧光也显示,G-iPSCs水凝胶培养后组织的牙周相关标志物呈强阳性表达。结论:作者的结果初步验证了G-和
