研究显示,内源性牙本质基质金属蛋白酶(MMPs)与牙本质粘接的树脂渗透层内胶原纤维自主降解相关联。为了靶向这些水解蛋白酶,我们有必要掌握在树脂/牙本质界面会激活和产生何种MMPs。在这篇研究中,作用从单颗拔除牙齿窝洞壁提取牙本质样本内源性MMPs的定量水平和提取程序进行优化。在作者构建的牙齿窝洞模型中,预备1个合面窝洞(2×4×2 mm)并从冠方去除,提取围绕周围的1-mm厚度牙本质壁。采用粉碎仪将样本制备成粉末状。酶联免疫吸附测定(ELISA)结果显示,在不到300 mg牙本质粉末中检测到平均34.7 pg的MMP-9。这是第一篇通过此种方法定量检测从单颗拔除牙齿窝洞壁分离出来的牙本质蛋白内的内源性MMP水平。原始出处:Kang J, Izutani N, et al.
