来自不同脑区的星形胶质细胞可以转化为不同的神经元亚型。

帕金森病的特征是大脑黑质部的多巴胺神经元的丧失。与其他主要的神经退行性疾病相似,至今为止,帕金森病没有任何改变疾病的治疗方法。

已有的大多数治疗策略的目的是防止神经元的损失或保护脆弱的神经元电路,此外,还有一个潜在的替代方案是取代失去的神经元,重建中断的脑回路。

最近,付向东团队报告了一个高效的一步转换法,通过耗尽RNA结合蛋白PTB(也称为PTBP1),其可以将分离得到的小鼠和人类星形胶质细胞转分化为有功能的神经元。

将这种方法应用于小鼠大脑,研究人员证明了星形胶质细胞逐步转化为新的神经元,支配到并重新填充到内源性神经回路。

来自不同脑区的星形胶质细胞可以转化为不同的神经元亚型。

使用化学诱导的小鼠帕金森病模型,研究人员发现,中脑星形胶质细胞会向多巴胺能神经元转化,多巴胺能神经元为重建黑质神经回路提供轴突。

值得注意的是,纹状体的再神经化伴随着多巴胺水平的恢复和运动障碍的改善。

通过使用反义寡核苷酸暂时抑制PTB,也可以将星形胶质细胞转化为神经元,并实现疾病表型的类似逆转。

这些发现确定了一种潜在的有效且临床可行的方法,通过替换丢失的神经元来治疗神经变性。
