Nature:警惕!高剂量的甜味剂三氯蔗糖会降低免疫反应

研究发现小鼠摄入高剂量的三氯蔗糖通过限制T细胞增殖和T细胞分化而产生免疫调节作用。

人工甜味剂在许多食品中被用作无卡路里的糖替代品,其消费量在过去几年里大幅增加。虽然一般被认为是安全的,但一些人对食用某些甜味剂的长期安全性提出了担忧。

2023年3月15日,英国弗朗西斯克里克研究所Fabio Zani等人在Nature在线发表题为“The dietary sweetener sucralose is a negative modulator of T cell-mediated responses”的研究论文,该研究发现小鼠摄入高剂量的三氯蔗糖通过限制T细胞增殖和T细胞分化而产生免疫调节作用。

在机制上,三氯蔗糖影响T细胞的膜顺序,同时降低T细胞受体信号传递和细胞内钙动员的效率。在皮下肿瘤模型和细菌感染模型中,给予三氯蔗糖的小鼠CD8+ T细胞抗原特异性反应降低,在T细胞介导的自身免疫模型中,T细胞功能降低。总的来说,这些研究结果表明大量摄入三氯蔗糖可以抑制T细胞介导的反应,这一效应可以用于缓解T细胞依赖性自身免疫性疾病的治疗。

另外,2022年9月7日,来自法国巴黎大学流行病学和统计研究中心的Charlotte Debras团队在国际知名医学期刊British Medical Journal(IF=93)杂志在线发表题为“Artificial sweeteners and risk of cardiovascular diseases: results from the prospective NutriNet-Santé cohort”的研究论文,这项大规模前瞻性队列研究结果表明,人造甜味剂的总摄入量与整体心血管疾病和脑血管疾病的风险增加有关。其中,阿斯巴甜的摄入与脑血管事件风险增加有关,而安赛蜜(乙酰磺胺酸钾)和三氯蔗糖与冠心病风险增加有关。总的来说,使用人造甜味剂代替添加的糖对心血管疾病没有任何好处。

2023年2月27日,美国克利夫兰诊所Stanley L. Hazen在Nature Medicine(IF=87)在线发表题为”The artificial sweetener erythritol and cardiovascular event risk“的研究论文,该研究检查了常用的糖替代品赤藓糖醇和动脉粥样硬化性血栓性疾病的风险。该研究结果表明赤藓糖醇既与MACE发病风险相关,又能促进血栓形成。研究评估赤藓糖醇的长期安全性是有必要的。

三氯蔗糖是一种常用的无卡路里甜味剂,其甜度约为蔗糖的600倍。尽管其吸收有限,但在人类食用含三氯蔗糖的食物或饮料后,可在循环中检测到三氯蔗糖,摄入250毫克三氯蔗糖可导致90-120分钟内血浆三氯蔗糖水平约为1 μM。人类每日可接受的最大三氯蔗糖摄入量(ADI)已由欧洲食品安全局(EFSA)确定为每公斤(体重)15毫克,或由美国食品和药物管理局(FDA)确定为每公斤(体重)5毫克。

基于体表面积(BSA)当量的异速缩放可用于根据小鼠代谢率的增加进行调整,将人类剂量的药物转换为小鼠剂量。通过让小鼠自由饮用含有0.72 mg ml?1或0.17 mg ml?1三氯蔗糖的水,研究人员计算出(使用BSA等量)10周内三氯蔗糖的消耗量接近欧洲食品安全局(0.72 mg ml?1剂量)或FDA (0.17 mg ml?1剂量)推荐的ADI当量。正如预期的那样,研究人员能够检测到与小鼠消耗增加相对应的循环三氯蔗糖的增加量,在最高剂量的三氯蔗糖下达到约1 μ M的血浆浓度,与人类可以达到的水平一致。
