FASEB J:发现DNA修复基因和男性不育有关!

为了进行这项实验,研究人员观察了一组野生型小鼠以及在原生殖细胞中有条件敲除XRCC1的小鼠。通过这两组小鼠,研究人员比较了XRCC1蛋白在睾丸中的表达、睾丸的大小和重量、体重、精子浓度和活力等。总的来说,研究表明XRCC1缺乏症会导致雄性小鼠因精子生成受损而不育。具体来说,研究人员发现,与野生型小鼠相比,敲除XRCC1的小鼠睾丸更小,精子浓度和活力也更低。

他们还证明,原生殖细胞中的XRCC1缺失与干细胞分裂、DNA损伤、氧化应激和凋亡有关。实验表明,N-乙酰半胱氨酸(NAC)治疗能够在一定程度上逆转上述几种情况,但不能恢复雄鼠的生育力或逆转XRCC1缺乏症引起的细胞凋亡。

“人类精子中轻微的DNA损伤就会降低生育能力,增加后代患病的风险,”南京医科大学公共卫生学院顾爱华教授解释说道。“现在我们知道XRCC1在维持小鼠正常精子生成中起着至关重要的作用,那么进一步的研究应该探索可能逆转XRCC1缺乏引起的男性不育的治疗方法。”

FASEB杂志主编Thoru Pederson博士说:“XRCC1一直被认为是许多细胞基因组完整性的守护者,这项新研究将其作用拓展到了精子形成的重要过程中。”
