自噬是细胞内物质的溶酶体依赖性降解过程,适应环境或发育变化,在维持细胞稳态中起着重要作用,而其功能障碍与多种人类疾病的发生相关。自噬的调节已经引起越来越多的关注,有望改善作为潜在治疗靶点的疾病的治疗。 近年来,金属有机骨架(MOFs)作为新兴的生物材料已经在生物学和生物医学领域进行了研究。因此,从安全性方面以及治疗观点来研究MOF对活细胞的影响是有意义且十分重要的,特别是它们对自噬的影响目前

近年来,金属有机骨架(MOFs)作为新兴的生物材料已经在生物学和生物医学领域进行了研究。因此,从安全性方面以及治疗观点来研究MOF对活细胞的影响是有意义且十分重要的,特别是它们对自噬的影响目前尚未报道。在这项研究中,研究人员探究了Fe-MIL-101_NH2对小鼠胚胎成纤维细胞(MEFs)的影响,并探讨了这些纳米粒子在自噬调节中的潜在应用。

结果表明,Fe-MIL-101_NH2诱导MEF中的细胞保护性自噬而无细胞毒性。自噬的激活使活性氧物质不会积聚,从而保护MEF免于凋亡。进一步探索MOF诱导的自噬的可能机制揭示了mTOR通路的抑制以及Becline1和Atg5的增强促成了自噬诱导。

综上所述,该研究揭示了MOF的自噬效应和机制见解,有益于MOF的安全性评估和合理有效的使用。
