骨肉瘤miRNA基因的差异性表达及其靶基因生物信息学研究

目的  筛查骨肉瘤miRNA(微小RNA)基因,建立骨肉瘤miRNA基因表达谱,分析明显上调或下调的基因。从中选取上调最为明显的基因和下调最为明显的基因来做进一步分析,验证基因芯片的结果。查找其靶基因并进行分析。
方法  提取7例骨肉瘤组织和正常骨组织的RNA,利用丹麦Exiqon公司的microRNA芯片对其进行分析。利用上一部分实验剩余的各组标本的total RNA,采用SYBR绿色荧光染料(SYBR GREEN )实时检测的逆转录一聚合酶链反应(RT-PCR)检测两种miRNA基因的表达,并用U6作为内参。利用TargetScan算法在因特网上的发布结果查询骨肉瘤miRNA基因的靶点。
结果  筛查出骨肉瘤中上调的基因有28个,下调的基因有26个;其中上调大于2.5倍的miRNA基因有5个:miR-381miR-18amiR-586miRPlus_42780miR-377*,表达差异最大的为miR-5865倍;其中下调大于2.5倍的miRNA基因有9个:miR-126miR-146amiR-16miR-191miR-142-5pmiR-106bmiR-144miR-26bmiR-20a,其中miR-144基因下调最为明显为39倍。标准曲线有良好的相关性,PCR产物特异。证实miR-586miR-144RT-PCR结果与芯片结果基本吻合。miR-586的靶基因目前发现有347个,考虑与骨肉瘤致病因素有关的基因为: TP53INP1, MTSS1, FGF5FGF9, TPT1等。miR-144的靶基因目前发现有647个,考虑与骨肉瘤致病因素有关的基因为: BCL11A, BCL11B, CDH11, TNFSF11等。
结论 建立了骨肉瘤的miRNA基因表达谱, miR-586miR-144基因为骨肉瘤表达最特异的基因。miR-586miR-144分别在骨肉瘤中明显上调和下调,可初步做为其特异基因。发现了一些miR-586miR-144有意义的靶基因,这些靶基因在促肿瘤作用中起到了直接或间接的作用。
