研究小组最初的工作是鉴定风险位点附近基因和调控区域的相关功能。这些位点不仅适用于评估此群体的乳腺癌风 险,也有助于了解疾病新的生物学特征。文章的资深作者,范德堡大学Ingram癌症中心的流行病学研究人员Wei Zheng认为,有必要开展进一步的机制研究,以便了解这些位点如何参与了乳腺癌的发生。

作为发现乳腺癌风险位点工作中的一部分,亚洲乳腺癌联盟(ABCC)汇集了亚洲各个国家过去十几项研究工作中的数据。总共,研究小组考虑了近23,000个东亚乳腺癌病例和超过24,000个群体匹配对照的信息。

在这项研究的发现阶段,研究人员直接比较了2,867名中国乳腺癌患者和2,285名中国对照的测定和估算 的SNP图谱,这些样本已利用Affymetrix的SNP 6.0芯片进行过基因分型。他们还评估了2,246名韩国乳腺癌患者和2,052名韩国对照,这些也利用相同的Affymetrix芯片分析过。

利用Illumina的Infinium芯片,研究小组对另外3,944名上海的乳腺癌患者和3,980名 对照开展分析,成功分型了约4,000个可疑的SNP位点,这些位点来自研究的第一阶段。此后,研究人员又对14,195个乳腺癌病例和16,249个对 照进行检测,对脱颖而出的几十个变异进行了分析,他们发现11个SNP仍与乳腺癌存在关联。

在整个病例和对照组的分析中,仅仅3个位点上的变异表现出全基因组的意义,它们分别是1号染色体ZC3H11A基因上的rs4951011、5号染色体ARRDC3基因上的rs10474352和15号染色体PRC1基因上的rs2290203。

在16,003个欧洲血统的乳腺癌病例和41,335个对照的后续分析中,研究小组追踪了相同位点上的变异,尽管关联较弱,但表明这些位点可能也导致其他群体的乳腺癌易感性。

来自ENCODE项目的数据表明,rs4951011和rs10474352变异可能分别位于增强子区域和转录因子结合位点。研究人员认为,这项研究的结果为乳腺癌的遗传学和生物学提供了更多的见解。
