Pharm Biol.:百合对对氯苯丙氨酸诱导的失眠症大鼠肠道菌群及代谢紊乱的缓解作用

失眠是世界难题,影响着所有年龄段的人。调查显示,全球总人口的30%受轻度失眠的影响,约10%患有严重或慢性失眠。失眠的原因和机制很复杂,可能是其他重大躯体疾病和精神疾病的症状,以及使用、滥用或暴露于某些物质。同时,越来越多研究证明,肠道微生物群会干扰宿主的胃肠道生理,代谢和免疫功能,并通过微生物群 - 肠道 - 大脑(MGB)轴影响中枢神经系统(CNS)功能和行为。肠道微生物群的微妙失衡与许多人类疾病有关,例如睡眠不足,昼夜节律错位,情感障碍和代谢疾病。

成人肠道中存在大约1,000种类型的微生物群,一项研究数据显示,体重70公斤的"标准人"有3.9×1013肠道中的细菌,细菌与人类细胞的比例为1:1。在另一项研究中,据报道,每个粪便样本中平均有762,655个肠道细菌基因,表明基因数量是人类基因组的38倍。上述数字突显了肠道菌群代谢能力的重要性。许多研究已经确定,肠道微生物群通过产生双向通信的各种途径影响大脑功能。最近的研究表明,生物钟错位和睡眠剥夺会改变生物钟基因表达和微生物群落结构。

使用中草药治疗失眠可以追溯到2000多年前。百合(LB)是一种常见的药用物种,该植物是中国龙徽县的地理标志产品。LB的鳞茎被称为"白鹤",白鹤连子汤、枸杞白鹤汤等以LB为主要原料的汤,在中国人中用于缓解失眠。在临床实践中,白鹤被中医生直接用作治疗失眠的中药,并记录在中华人民共和国药典中(Munafo and Gianfagna

LB在临床上已被长期用于治疗失眠,但其机制仍未得到探索。本研究采用腹腔内(ip)PCPA诱导的失眠大鼠模型。检测失眠大鼠下丘脑相关神经递质、对应受体和病理,采用16S核糖体RNA(16S rRNA)基因测序联合超高效液相色谱-质谱/质谱(UPLC-MS/MS)综合分析LB对PCPA诱导失眠大鼠肠道微生物群和粪便代谢表型的影响。

在Wistar大鼠腹膜内注射对氯苯丙氨酸(PCPA)诱导失眠模型。将大鼠分为3组:对照组、PCPA (400 mg/kg,灌胃2天)、LB(598.64 mg/kg,灌胃7天)。观察下丘脑5-羟色胺(5-HT)、去甲肾上腺素(NE)、褪黑素(MT)水平、GABAA、5-HT1A、MT受体的表达及病理变化。采用16S rDNA测序和UPLC-MS/MS技术检测肠道菌群和代谢变化。

1、LB对PCPA诱导失眠大鼠体重的影响

结果显示,对照组和PCPA组大鼠的体重变化存在差异(图1)。与PCPA组相比,LB组的体重增加显著增加。

2、LB对下丘脑5-HT、NE和MT的影响

图2(A-C)所示,PCPA给药显著降低了下丘脑的5-HT和MT水平(p <0.05),并增加了NE的水平(p<0.01)。每天口服LB(598.64mg / kg)7天,使5-HT和MT水平显着增加(p <0.01)和NE水平降低。

3、组织病理学观察

图3(A-C) 所示,对照组下丘脑神经细胞不仅丰富且形状良好,而且分布均匀,而PCPA组的神经细胞严重变形,排列不紧密,并且有许多破碎的小细胞。值得注意的是,在LB组中,细胞排列整齐,萎缩细胞的数量显着减少。

4、LB对下丘脑GABAA R、5-HT1A和MT表达的影响

如图4(A-C)所示,PCPA处理显著减少了下丘脑5-HT和GABAA R的表达(p <0.01),但MT的表达无显著变化。口服LB (598.64 mg/kg)超过7天,显著的增加 5-HT1A、GABAA R和MT的表达(p <0.01)。

5、粪便中的细菌群落组成

我们对16S rRNA基因的V3-V4可变区进行了测序。以97%的序列相似度为临界值,将高质量序列聚类为51,342个操作分类单元(operational taxonomic units, OTUs)在LB组中,OTU秩丰度在横轴上的斜率较平缓,分布较宽,说明添加LB后肠道菌群更加多样化,分布更加均匀(图5(A))。同时,接近平台的稀疏曲线表明测序深度含有罕见的新系统型和大多数多样性(图5(B,C))。Chao1的估计值反映了细菌群落的生态物种丰富度,突出了物种数量,而Shannon指数与物种分布呈正相关。Chao1指数平均为3539.91比2901.54(p <0.01)(图5(B)),Shannon指数平均值为7.84 vs. 7.11(p<0.05)(图5(C)), LB组均高于PCPA组。

6、LB对PCPA诱导失眠大鼠肠道菌群的影响

在OTU水平上,计算了两组样本之间的差异和相似性(图6(A))。与PCPA组(4725)菌群多样性的降低相比,在LB添加后植株多样性显著增加(5220),趋向于正常组(5324)。PCoA分析表明,LB组的群落组成与PCPA组分离(图6(B))。

分类单元总结揭示了LB干预后肠道微生物组成的显著变化。在属水平上,10个区系在PCPA组与对照组之间存在显著差异(图6(C))。与对照组相比,PCPA组紫单胞菌科(18.92%,p <0.01),瘤胃球菌科(6.46%,p <0.01),梭状芽胞杆菌(1.35%,p<0.05)的百分比显着下降,而乳酸菌(14.02%,p <0.01),普氏菌(7.85%,p <0.05),大肠埃希菌(8.15%,p <0.01),拟杆菌(1.81%,p <0.01)和萨特氏菌(1.26%,p <0.05),布劳特氏菌(1.06%,p <0.05),副拟杆菌(0.81%,p<0.05)显着增加。 同时,5个菌群在属水平上LB组与PCPA组之间存在显著差异(图6(C))。与PCPA组相比,LB组紫单胞菌(28.83%,p<0.05)、毛螺菌(14.06%,p<0.05)和罗氏菌(4.26%,p<0.01)的百分比显著增加,而乳酸菌(6.13%,p<0.01)和大肠埃希菌(1.27%,p <0.01)的比例明显下降。

7、LB对PCPA诱导失眠大鼠粪便代谢组学分析的影响

根据OPLS-DA模型,LB组的代谢特征与PCPA组的代谢特征有显著差异,但往往与对照组相似,表明PCPA引起的改变在LB添加后明显缓解。在PCPA和LB组中差异表达的代谢物以火山图(图7(C,D))显示。

为了鉴定PCPA和LB组之间的代谢产物差异,我们选择了PCPA和LB组中变化倍数为≥1.5(上调)或≤0.5 (下调)的代谢产物。这些从OPLS-DA模型中筛选出VIP≥1的代谢产物。最后从所有样本中筛选出662个代谢产物(图7(C,D))。其中,131个代谢物在PCPA处理后上调,65个代谢物下调(图7(C))。与PCPA组相比,LB组有33个代谢产物表达上调,65个代谢产物表达下调(图7(D))。这些代谢物被分为20多个不同的类,但主要是有机酸及其衍生物、核苷酸及其代谢物、氨基酸及其代谢物、氧化脂类、碳水合物及其代谢物、脂类其他磷脂、苯及其取代衍生物、酚类及其衍生物、脂类等。如图7(E、F)所示,在PCPA和LB组中,前10种差异代谢物分别呈上调(红色标记)和下调(绿色标记)。在这些总差异代谢物中,与PCPA组相比,LB组中共有37种代谢物显着下调或上调(表1)。

8、差异代谢物的KEGG分类和富集分析

随后使用KEGG对所有差异代谢物进行了富集分析。研究表明,PCPA诱导的代谢产物主要与花生四烯酸代谢、色氨酸代谢、原代胆汁酸生物合成、不饱和脂肪酸生物合成、血小板活化等有关(图8(A))。与PCPA组相比,LB改变的差异代谢物主要与抗叶酸耐药性、花生四烯酸代谢、嘌呤代谢、色氨酸代谢、TRP通道的炎症介质调节等有关(图8(B))。

9、粪便微生物群与代谢物的相关性分析

为了深入了解肠道微生物群的分布是否与代谢特征相关,我们使用Spearman相关性分析分析了显着不同的属级菌群和差异代谢物之间的相关性(|r|> 0.7)。如热图(图 9)所示,总共 33 种差异代谢物与 LB 组中 28 种肠道菌群中的一种或多种呈正相关或负相关。其中,LB处理后趋向正常的代谢产物有(±)9-HETE,(±)12-HETE,15-氧代ETE,1-甲基尿酸,2-氨基己二酸,8,9-EET,生物喋呤,d-果糖,DHA,犬尿酸,l-肉碱,l-棕泡蝶呤,泛醇,磺胺嘧啶,色胺,黄嘌呤。

综上,LB逆转了大鼠肠道菌群和粪便代谢表型多度和多样性的不良变化,并且改善了下丘脑细胞的病理现象。PCPA改变最显著的花生四烯酸代谢和色氨酸代谢途径受到LB的显著调控。此外,还发现LB降低了与精神疾病相关的犬尿酸和与心血管疾病相关的三甲胺-N-氧化物的水平。

总之,LB可以通过改善PCPA引起的肠道菌群和代谢物的紊乱来缓解失眠,作为一种药用和食用草药,LB可以考虑作为保健食品进行开发,以缓解未来日益增加的失眠症。
