B细胞AIM2缺陷可通过调节 Blimp-1-Bcl-6介导的B细胞分化改善系统性红斑狼疮

该研究发现AIM2在狼疮患者的B细胞中高表达,并通过调节Bcl-6–Blimp-1促进B细胞分化,为系统性红斑狼疮治疗提供了新的靶点。

    据报道AIM2是先天免疫细胞炎症小体的组成部分。令人惊讶的是B细胞表达AIM2,且在狼疮患者的B细胞中观察到更高的AIM2表达。迄今为止,B细胞中独立于炎症小体的AIM2功能尚不清楚。

     这篇来自中南大学湘雅附属第二医院Dr. Qianjin Lu团队的研究报告了AIM2在人扁桃体的记忆B细胞和生发中心 (GC) B细胞的表达增加,在狼疮患者的循环记忆B细胞和浆细胞、以及皮肤中均表达增加。采用CD19 Cre小鼠模型在B细胞中条件敲除AIM2降低了淋巴结和脾脏中的CD19+ B细胞频率,并抑制了KLH诱导的IgG1抗体产生。在Pristane诱导的狼疮小鼠模型中,B细胞中的AIM2 缺乏会减轻狼疮症状并降低GC B细胞、滤泡辅助T细胞(Tfh)、浆母细胞和浆细胞的频率。此外,人B细胞AIM2缺失使Blimp-1表达增加并降低了Bcl-6表达。然而沉默Blimp-1Bcl-6AIM2表达没有显著影响,表明AIM2可能是Blimp-1Bcl-6的上游调节器并通过Co-IP实验证明AIM2直接与Blimp-1Bcl-6结合。此外,发现IL-10通过DNA去甲基化上调AIM2表达。

结论:

     该研究发现AIM2在狼疮患者的B细胞中高表达,并通过调节Bcl-6Blimp-1促进B细胞分化,系统性红斑狼疮治疗提供了新的靶点。
