PLoS Biol:少吃肉,记忆好!中国科学家发现饮食限制,尤其是少吃含色氨酸食物,增强记忆力的神经机制,考前吃素或能考更好

近日,Plos Biology杂志上发表了一项来自台湾国立大学王培育副教授课题组的研究成果,研究者们发现了少吃有助记忆力的神经通路,原来关键在于各种蛋白质中含有的色氨酸!减少色氨酸摄入会减少5-羟色胺生物合成,影响神经元的可塑性,增强记忆力。

曾经,有这样一个问题摆在奇点糕面前:有钱但丑,和贫穷但好看,选哪个?

而今天的问题恐怕更加触及生理需求底限:吃不饱但聪明,和有肉吃但记性差,选哪个?

饮食限制(DR),即在保证营养的情况下,减少每日摄入的总热量20-40%,被认为是目前最有效的抗老措施,不仅可以改善老年人的认知,还成功在几种模式动物中实现了延长健康寿命的神奇效果。

少吃饭就这么神奇?难道真的是“人这辈子九吨饭,谁先吃完谁先散”?

研究者找来了两组小鼠,一组只有4个月大年轻力壮,一组则已经24个月到了风烛残年。它们再分两组接受正常饮食(AL)和60%热量的限制饮食(DR)。

然后研究者给小鼠做了个新目标识别测试(NOR),目的是测量小鼠的短期记忆(1小时)和长期记忆(24小时)。结果特别明显,无论年轻年老,小鼠的短期记忆能力都差不多,但吃得少的明显长期记忆力更好!

难道是小鼠一直吃不饱,饥肠辘辘才注意力更加集中?

不然。研究者也试着先让正常饮食小鼠饿18个小时,或提前2小时给限制饮食小鼠吃顿饱饭,结果还是一样的,长期限制饮食的小鼠记忆力还是比较好的。

有意思的是,想要拥有更好的记忆力,得持续少吃至少两个星期才行,如果让少吃小鼠再回到自由吃饭的生活,过两周它们就又“记不住事儿”了。

看来应该是有某种营养成分的缺失起了作用。研究者分别设计了限制饮食+单独碳水化合物/蛋白质/脂肪的食谱,推断出秘密藏在蛋白质里。

组成蛋白质的各种氨基酸确实也比较可疑。比如说谷氨酸,它自己就是一种神经递质;又比如说色氨酸、酪氨酸和半胱氨酸,它们分别是5-羟色胺、儿茶酚胺和含硫氨基酸的前体物质,这些物质都是可以调节认知功能的。

再次控制变量法,幕后boss找到了,原来是色氨酸!

如果沿着色氨酸继续往下探索,恐怕5-羟色胺跑不掉。研究者提前30分钟给小鼠注射了芬氟拉明,这是一种促5-羟色胺释放剂,可提高5-羟色胺水平。果然,这批小鼠靠少吃得来的好记忆力,又回去了。

可见,应该就是少吃→少色氨酸→少5-羟色胺→记性好这么一条线了。

接下来就是寻找具体的作用通路了。研究者对5-羟色胺的合成、代谢和它的一系列受体进行了监控,发现少吃小鼠的海马和其他与记忆有关的脑区中,5-羟色胺受体6(HTR6)表达减少了很多。

用药物激活HTR6可以抵消少吃带来的好记忆,拮抗HTR6或干脆敲除它则能够让正常吃饭小鼠记忆变好。

这也排除了少吃本身的影响。以前曾有研究证实,限制饮食或间歇性进食带来的外周的葡萄糖代谢或酮生成,与寿命延长和神经可塑性有关。


用药激活HTR6记忆力就回去了(左)

在HTR6的下游,则是我们熟悉的老朋友mTORC1通路,激活mTORC1也同样抵消了少吃小鼠的好记忆。如果用雷帕霉素类似物拮抗mTORC1,则能够模拟少吃的作用。

说起来雷帕霉素也的确是少数被证实可以提高记忆力、延长寿命的分子之一。

至于少吃之后,神经元到底发生了些什么变化,研究者也做了一番分析。结果显示,神经元的树突复杂程度下降了,长度也缩短了,大脑神经元密度虽然没变,但脊柱神经密度增加了,而且海马的神经元长时程增强(LTP)也增加了。

LTP指的就是两个神经元信号传输中的一种长时间增强现象,是够成学习和记忆基础的机制之一。


整个作用通路差不多就是这样

奇点糕以前写过另一篇文章,聪明人的大脑反而更“简洁”,神经元之间的连接更少,但是数量更多。万万没想到这种智慧可能来源于少吃肉……
