PNAS:日本学者建立新质粒反向遗传学系统

轮状病毒是引发严重腹泻的最常见原因之一,每年都会夺去数十万婴儿的性命,虽然目前的疫苗能有效预防轮状病毒的恶化,然而目前的技术手段无法分析轮状病毒如何在细胞中侵入和复制的。

Takeshi Kobayashi说,“反向遗传学能帮助我们获得人工改造的病毒。使用反向遗传学,我们可以令一个基因发生突变,然后观察这对病毒的影响”。

目前反向遗传学已经被用于了许多种病毒研究中,用以分析病毒繁殖的条件,但是要针对像是轮状病毒这样的多片段RNA病毒细胞并不容易,这一研究组通过两个病毒蛋白:FAST和VV capping enzyme解决了这个问题,他们将这两种蛋白加入到质粒系统中,利用其优势,突变了轮状病毒的单个蛋白:NSP1,结果表明这可以减少病毒的复制,从而检验了这一系统。

未来通过进一步在所有蛋白中进行全面检测,研究人员希望能找到令轮状病毒成为严重公共威胁的关键决定因子,“我们希望可以修改轮状病毒的繁殖和致病性,”Kobayashi说。

Kobayashi对于这一质粒反向遗传学系统充满信心,“因为没有人可以人工合成轮状病毒,因此我们对这种病毒的复制和发病机制了解甚少”。他希望这一新系统将促进轮状病毒的研究工作,并指导研发更有效的疫苗。
