摘要:

造血干细胞移植(HSCT)常被用来治疗血液病。供体HSCs必须在重建生态位的破坏环境中存活、增殖和分化。阐明移植HSCs活性的分子机制将有助于改善HSCT的预后。

Gprasp1或Gprasp2基因沉默可增加移植后造血干细胞和祖细胞(HSPCs)的存活、静止、迁移、壁龛保留和造血再生活性。

研究人员进一步证明GPRASP1和GPRASP2可促进移植过程中HSC功能的主要调节因子CXCR4的降解。CXCR4在缺乏Gpras的HSPCs中积累,增强了HSPCs移植后的功能。因此,GPRASPs负调控HSCs中CXCR4的稳定性。

本研究表明,GPRASP蛋白是HSCT和CXCR4活性的负调控因子。将来或可通过打断GPRASP和CXCR4的相互作用的来提高HSCT的效果。
