Redox Biol:自噬途径是预防衰老相关骨病的潜在作用靶点

细胞自噬(autophagy)是真核生物保守的对细胞内物质进行周转的重要过程。

细胞自噬(autophagy)是真核生物保守的对细胞内物质进行周转的重要过程。该过程中一些损坏的蛋白或细胞器被双层膜结构的自噬小泡包裹后,送入溶酶体或液泡中进行降解并得以循环利用。机械负荷和细胞内自噬在骨稳态中起重要作用,但它们之间的关系仍然未知。

研究人员调查了骨细胞是否在流体剪应力(FSS)加载时发生自噬,以及机械诱导的自噬和ATP代谢之间的相关性。研究人员通过透射电子显微镜(TEM)观察在FSS作用下的骨样细胞样MLO-Y4细胞中的自噬泡以及FSS处理后的GFP-LC3转化细胞。

结果显示,通过抑制3-MA自噬能够减弱骨细胞中FSS诱导的ATP释放和合成。在3-MA(自噬抑制剂)预处理的培养物中暴露FSS后,能够观察到高比例的细胞死亡,但Caspase3/7的活性没有发生显著的变化。结果表明FSS能够诱导骨细胞发生保护性自噬,并且机械诱导的自噬与ATP代谢和骨细胞存活相关。LC3-Ⅱ亚型的增加和p62的降解证实了FSS加载会增加自噬流量。

该研究表明,用于调节自噬状态的药物可以提高骨骼细胞的存活率,自噬相关的途径可能成为预防衰老相关骨病的作用靶点。
