干细胞疗法一直被认为是治疗许多疾病的理想疗法之一。然而,这种疗法潜在的副作用是制约这种疗法发展的重要原因之一。作为一种再生医学疗法,干细胞疗法主要是通过向患者体内输送干细胞,使其来分化成为具有正常结构、功能的细胞或组织以达到替换病变部位的效果。然而,这一疗法的风险之处在于输送至体内的干细胞一旦脱离控制可能会发育成其他类型细胞,从而失去治疗效果。另一方面,输入的干细胞还可能会引起患者体内免疫反

最近,来自美国北卡罗来纳州和中国郑州的研究人员就独辟蹊径开启了一个新的研究思路。研究人员利用PLGA纳米粒来模拟细胞结构,并在这个微粒上修饰从人心肌干细胞中提取的生长因子蛋白,最后再在这一结构表面覆盖上一层心脏干细胞膜,从而构建出了一种全新的“人造心肌干细胞”。

体外实验显示,这种“人造心肌干细胞”与真正的心肌干细胞一样都可以促进心肌细胞的分化和生长。在心脏病动物模型上,这种结构同样促进了新的心肌组织的形成。

研究热暖表示,这意味着这种“人造心肌干细胞”刺激心肌细胞发育的机制类似于传统的减毒疫苗。最外层的细胞膜可以保证其逃避机体免疫系统并结合到心脏部位,通过释放内部的细胞因子达到刺激组织细胞再生。同时,这种颗粒无法像真正的干细胞那样扩增,因而有效避免了干细胞疗法潜在风险。

此外,这种“人造心肌干细胞”在临床生产中有着更大的优势。生产人员可以很容易的进行冻干和密封操作,大大延长了其保存期。

研究人员表示,这一方法是开发一种新型常备型干细胞疗法产品的第一步,有可能能最大限度的发挥干细胞疗法的优越性。
