Int J Mol Sci:三维石墨烯-RGD肽纳米复合材料增强人脂肪源性间充质干细胞的成骨作用

将胺修饰的二氧化硅纳米粒子(SiNPs)均匀涂布在氧化铟锡电极(ITO)上,随后进行氧化石墨烯(GO)封装和金纳米粒子的电化学沉积。RGD-MAP-C肽具有三重分支的重复RGD序列和末端半胱氨酸,自组装到金纳米颗粒上,产生最终的三维石墨烯-RGD肽纳米岛复合物。我们利用各种金纳米粒子-RGD肽簇密度生成了底物,具有最大数量簇的复合物最适合于ADSC粘附和扩散。

值得注意的是,基于基因表达(碱性磷酸酶(ALP),runt相关转录因子2),酶活性(ALP)和钙沉积,相同复合物在指导ADSC成骨方面与其他底物相比也非常有效。诱导分化为成骨细胞的ADSC在14-21天后表现出比在典型的GO-SiNP复合物上生长更高的钙积累,表明该复合物可加速ADSC成骨细胞分化。

结果表明,三维石墨烯-RGD肽纳米岛复合物可以有效地从间充质干细胞衍生成骨细胞。
