转移导致90%的癌症患者死亡。癌症通过转移扩散是由一系列称为癌基因的突变蛋白驱动的,这些癌基因导致癌细胞不受控地繁殖并促进其移动能力。癌基因活性如何特异性地指导增加的运动和转移是非常复杂的并且基本上未知。

UH癌症中心副主任Joe W. Ramos博士和合作者集中研究这些癌基因和相关信号如何导致细胞内正常过程失调,并激活高度移动的和侵入性的癌细胞行为。

这一发现发表在“美国科学院学报”(PNAS)上,定义了癌基因打开癌细胞移动所需的蛋白质RSK2的机制。ARamos及其同事发现,RSK2蛋白形成了一个信号中枢,包括名为LARG和RhoA的蛋白。他们表明,打开这个信号枢纽激活了癌细胞的运动。这些结果显著提高了癌细胞在转移期间如何移动的理解,并且可以提供更精确的药物靶标以阻止具有致癌突变的患者的癌症转移。

“这些新的数据是非常令人兴奋的,阻止癌症侵袭和转移仍然是治疗患者的核心挑战,我们预计这一研究可能会导致脑肿瘤,黑色素瘤和乳腺癌等新的治疗机会,目前我们正在关注这些机会和开发新的化合物来瞄准这个信号枢纽,”拉莫斯说。
