由德累斯顿工业大学的Michele Solimena教授,法兰克福的Sanofi的Anke M Schulte博士,瑞士生物信息学研究所(洛桑)的Mark Ibberson博士和比萨大学的Piero Marchetti教授欧盟创新医药行动(IMI)研究联盟IMIDIA已经在2型糖尿病患者的胰岛中发现了一组新的失调基因。这些发现现在发表在Diabetologia(欧洲糖尿病研究协会(EASD)杂志)上。

从2010年2月2日至2016年9月,涉及14家欧洲学术机构,大型制药公司和生物技术公司的IMIDIA联盟的目标是确定胰岛素生产型胰岛β细胞的再生,维持和保护的新途径,以加速发现更有效的预防和治疗糖尿病的策略。 IMIDIA联盟的主要任务是确定哪些基因在糖尿病受试者的胰岛β细胞中异常表达,与非糖尿病受试者的胰岛β细胞相比。这些基因的改变的表达可以解释糖尿病中的β细胞衰竭。研究人员首次基于他们的比较性基因表达分析,不仅从非糖尿病和糖尿病器官供体(其临床信息的可用性是有限的)收集的胰岛,而且还从接受胰腺手术的患者的胰岛中收集胰岛。对于后者的深入医疗信息是可用的,糖尿病状态可以在手术前准确评估。

因此,这种方法能够组装来自非糖尿病和糖尿病受试者的最大量的胰岛集合,还能够获得来自糖尿病前期个体的胰岛。研究人员通过这种方式鉴定了与来自这些群体的非糖尿病胰岛相比,在糖尿病器官供体以及糖尿病手术患者的胰岛中表达改变的19种基因。值得注意的是,这19个基因中的9个以前从未显示在糖尿病胰岛中失调。另一方面,研究未发现任何这些基因在前驱糖尿病受试者的胰岛中失调的证据,因此表明其改变的表达是糖尿病中β细胞失败的原因而非结果。因此,未来的研究将致力于发现哪些基因在β细胞消亡之前失调。为此,必须从前驱糖尿病受试者那里收集更多的胰岛,这正是最近启动的欧盟 - 伊米联盟RHAPSODY所追求的主要任务之一,其中包括所有四个领导本次IMIDIA研究的团队作为合作伙伴。
