Blood:Ly6C-单核细胞存活依赖C/EBPβ。

摘要:

转录因子CCAAT/增强子结合蛋白β(C/EBPβ)在单核细胞/巨噬细胞中高表达。但其在单核细胞中的作用基本尚不清楚。故此,研究人员对C/EBPβ在单核细胞中的作用进行研究。

对单核细胞进一步细分发现Cebpb mRNA在骨髓Ly6C-单核细胞中的表达显著上调。相应的,Ly6C-单核细胞在Cebpb-小鼠中明显减少。骨髓嵌合体实验和Mx1-Cre-介导敲除Cebpb实验揭示在单核细胞发生过程中,细胞内源性和单核细胞特异性的对C/EBPβ的需求。

在Cebpb-/-小鼠中,Ly6C-单核细胞的周转率显着加速,其凋亡也增加。编码巨噬细胞集落刺激因子的Csf1r,在Cebpb-/-小鼠的Ly6C-单核细胞中的表达明显下调。C/EBPβ结合Csf1r的正向调控元件,促进其转录。

总之,以上结果表明C/EBPβ是Ly6C-单核细胞的存活的一个关键因子。
