Int J Mol Sci:长期冷冻保存的牙髓干细胞可在体外分化成胆碱能神经元并在体内移植时增强运动神经的再生

在体外分化期间,细胞显示出神经元样形态变化,包括轴突纤维和神经元体发育,并且在信使RNA(mRNA)和蛋白质水平上显示出胆碱能神经元特异性标志物的高表达。重要的是,DF-chN具有Ach分泌能力。在具有坐骨神经缺损的大鼠中DF-chN移植后8周,通过开放场测试检测到行为活动明显增加,使用免疫组织化学检测增强的低亲和力神经生长因子受体(p75NGFR)的表达。

综上所述,该研究结果表明,来自冷冻保存的牙髓的干细胞可以在体外成功分化成胆碱能神经元并在体内移植时增强运动神经再生。此外,该研究表明,长期保存的牙髓组织值得用作神经再生领域的自体细胞资源,包括胆碱能神经。
