Diabetologia:腺嘌呤核苷酸对2型糖尿病小鼠肝脏PTP1B活性的调节作用

本研究采用高效液相色谱法测定血浆和肝脏中腺嘌呤核苷酸及其代谢产物。采用1H NMR为基础的代谢组学分析,探讨2糖尿病小鼠模型中代谢物的变化。对腺嘌呤核苷酸代谢产物进行Na+/K+ ATP酶和Na+/H+交换活性测定。采用重组蛋白酪氨酸磷酸酶1B(PTP1B)进行酶动力学测定。采用微量热电泳法进行蛋白质结合试验。用BCECF-AM法测定肝细胞AML 12细胞内pH值。此外,还分析了2型糖尿病患者的PAMP水平。

研究发现pAMP升高是所有2型糖尿病小鼠模型的普遍现象,包括db / db与瘦小鼠(13.9±±2.3μmol/ l3.7±±0.9μmol/ l; p <0.01),ob / ob与瘦小鼠(9.1 ±2.0μmol/ l3.9±1.2μmol/ l; p <0.01)和高脂饮食/链脲佐菌素诱导的野生型小鼠(6.6±1.5μmol/ l4.1±0.9μmol/ l; p <0.05 ;这种升高是肥胖小鼠中发生高血糖所必需的。 HPLC分析后的基于1H NMR的代谢组学研究表明,经5'-AMP处理的野生型小鼠的代谢产物谱与db / db糖尿病小鼠相似,尤其是大量ATP及其代谢产物的积累。降糖药物二甲双胍可降低5'-AMP诱导的野生型小鼠和db / db小鼠的高血糖严重程度。在这些高血糖小鼠中,二甲双胍降低了肝脏ATP的积累,但没有降低其代谢产物。 ATP和二甲双胍会相应地改变肝脏中细胞的pH稳态,从而导致PTP1B(胰岛素信号的关键负调节剂)的肝脏活性发生相反的变化。此外,pAMP水平在2型糖尿病患者中也有所升高(45.2±±22.7 nmol / l3.1±±1.9 nmol / l p < 0.01)。

这些结果揭示了腺嘌呤核苷酸在调节高血糖中的新作用,并为肥胖和2型糖尿病提供了一个潜在的治疗靶点。
