Nature:孙颖浩教授研究组全新发布亚洲人前列腺癌基因组图谱

通过对2554例前列腺肿瘤的系统比较,绘制了亚洲人群前列腺癌基因图谱,研究表明中国患者的基因组突变特征与西方患者明显不同。

前列腺癌是世界范围内高发和主要的致死癌症之一。在中国,2015年约有60300个新增前列腺癌患者和26600个前列腺癌死亡患者,该数据从2000年起,就以每年12.6%的比率不断攀升。

近日,来自第二军医大学长海医院的孙颖浩教授等人领导的研究组发表了题为“A genomic and epigenomic atlas of prostate cancer in Asian populations”的文章,通过对2554例前列腺肿瘤的系统比较,绘制了亚洲人群前列腺癌基因图谱,研究表明中国患者的基因组突变特征与西方患者明显不同。

这一成果以“Article”的形式公布在3月25日的Nature杂志上。

孙颖浩教授长期致力于前列腺癌、泌尿系结石和微创泌尿外科技术的研究,并取得不少成果,此前生物通也陆续报道了相关一些成果。2017年,这一研究组首次完成了对亚洲前列腺癌患者的大规模基因组测序工作。

在此基础上,在最新这篇文章中,研究人员通过与已发表的欧美人前列腺癌基因组进行对比,发现在亚洲人群中,除了存在已知的显着突变基因SPOP、TP53和PTEN等外,还存在高频的CHD1基因缺失以及AR通路上游调控基因的突变。

研究人员分析了来自中国原发性前列腺癌患者的208份肿瘤组织样品和匹配的健康对照组织的全基因组,转录组和DNA甲基化数据。

他们发现,中国患者的基因组改变特征与西方人群明显不同:具体而言,41%的肿瘤包含FOXA1突变,18%的ZNF292和CHD1缺失。基因组和表观基因组的变化是相关的,并且可以预测疾病的表型和进展。编码和非编码突变都集中在对前列腺癌重要的途径上,从而为这种毁灭性疾病提供了见识。

研究人员表示,这些发现强调了在构建疾病的综合基因组图谱时考虑人群背景的重要性,揭示了东西方人前列腺癌基因组图谱的巨大差异,发现了与前列腺癌预后及生存相关的新基因,将为前列腺癌的治疗、诊断和预后检测提供新的基础。
