外泌体是在许多生物过程中发挥重要作用的纳米级细胞外囊泡,包括细胞间通讯、抗原呈递以及蛋白质、RNA和其他分子的转运。最近,人们对外泌体相关的基础研究产生了极大的兴趣,寻求用于健康监测和疾病诊断的新的基于外泌体的生物标志物。

本研究中,研究人员报告了基于声学流体(即声学和微流体的整合)的分离方法,以无标记和无接触的方式从全血中直接分离外泌体。该声学流体平台由两个模块组成:一个微型细胞去除模块,首先去除较大的血液成分,然后在外泌体分离模块中进行细胞外囊泡亚组分离。

在细胞去除模块中,研究人员证明从微米和纳米尺寸颗粒的混合物中分离110纳米颗粒,产率大于99%。在外泌体分离模块中,研究人员从胞外囊泡混合物中分离外来体,纯度为98.4%。将两种声学流体模块集成到单个芯片上,从全血中分离出外泌体,血细胞去除率超过99.999%。

综上所述,该研究结果表明,集成的声学流体装置能够以无标记、无接触和连续流动性的方式快速分离外泌体,且生物相容性良好,为探究外泌体在人类疾病发生和发展中、健康监测、医疗诊断、靶向给药和个性化医疗中外泌体的分离提供了新的简便且高效的方法。
