乙型脑炎患者的死亡率较高,且存活患者易患神经性后遗症。因此,研发针对乙型脑炎病毒的特异药物和新疫苗对该流行病的防治具有极其重要的意义。2F2 和 2H4 这两株中和抗体是 30 年前第四军医大学微生物学教研室筛选得到的高中和活性的抗体,在当时乙型脑炎经常性爆发的特殊时期曾用于患者的临床治疗,且表现出很好的安全性和治疗效果。

在研究中,研究人员首先发现 2F2 和 2H4 这两株中和性抗体只与乙型脑炎病毒相互作用,不与同为黄病毒属的登革热病毒、寨卡病毒和黄热病毒相互作用,并且这两株抗体均表现出了较高的亲和力。通过细胞水平的中和实验发现,这两株抗体均具有极好的中和活性。动物水平的保护实验发现,这两株抗体能够完全清除被感染小鼠脑内的乙型脑炎病毒,并完全治愈被感染小鼠。

研究人员通过冷冻电镜的方法解析得到 4.7 ? JEV-2F2 和 4.6 ? JEV-2H4 的复合物结构。通过结构分析发现,黄病毒属中具有较高中和活性的抗体均结合在 E 蛋白的四级结构上,且这些抗体均为病毒特异的中和性抗体。因此可以根据以上特点,设计针对病毒的特异性新型疫苗。通过进一步研究两株中和性抗体的作用机制,研究人员发现这两株抗体不仅能阻止病毒和细胞的结合,还能阻止病毒和细胞在酸性条件下的融合,并且起主导作用。

该研究得到国家自然科学基金、科技部 973 项目,中科院先导专项等资助,同时得到军事医学研究院研究员秦成峰和俄亥俄州立大学研究员刘善虑的支持。中国科学院生物物理研究所研究员王祥喜、空军军医大学教授徐志凯与中科院院士饶子和同为本文的共同通讯作者,饶子和团队博士生邱肖迪和空军军医大学副教授雷迎峰为本文共同第一作者。
