Oncogenesis:长期接触镉能够诱导良性前列腺上皮细胞的恶性转化

最近,有研究人员确定了是否Cd接触能够恶性转化BPH1细胞以及探索了潜在的机制。研究通过RNA测序解析了与BPH1转化有关的分子信号,并发现Cd能够诱导小脑锌指2(ZIC2)在BPH1细胞中的表达。研究人员也注意到了Cd接触能够增加ZIC2在Cd转化的BPH1细胞中的表达,从而反过来促进非锚定球体和增加干细胞驱动因子的表达,表明了其在干细胞自我更新中的作用。转化的细胞中ZIC2表达的沉默能够抑制球体的形成、干细胞标记的表达和无毛鼠中肿瘤的生长。在分子水平,ZIC2能够与glioma相关的致癌基因家族(GLI)锌指1(GLI1)互作,从而激活Cd接触BPH1细胞中的促生存因素信号。与之相反,ZIC2在BPH1细胞中的过表达能够引起球体的形成,从而确定了ZIC2的致瘤功能。在原代BPH细胞中Cd接触诱导的ZIC激活和GLI1信号确定了上述致瘤功能的临床意义。最后,人类BPH样本与临近健康组织相比ZIC2表达增加。

最后,研究人员指出,他们报道了Cd接触能够通过ZIC2和GLI1信号的激活来诱导BPH恶性转化的直接证据。
