Blood:靶向骨髓壁龛可挽救β-地中海贫血受损的造血干细胞功能

β-地中海贫血的HSC自我更新能力受损,是因为持续处于改变的BM微环境中所造成的。 纠正HSC-基质壁龛交联可通过体内再激活PTH信号恢复地中海贫血的HSC功能。

造血干细胞(HSCs)是由骨髓(BM)壁龛的信号调控的,在稳定状态下和血液疾病中调节血细胞生成。为了了解HSC-壁龛相互作用在非恶性的改变的稳态中的作用,研究人员选择了一种血红蛋白病——β-地中海贫血作为范例入手。

在这种严重的先天性贫血中,原发性血红蛋白缺陷的继发性改变对HSC-壁龛交联具有潜在的影响。在本研究中,研究人员发现地中海贫血小鼠(th3)的造血干细胞由于BM壁龛相互作用改变而导致功能受损。

HSC的自我更新缺陷在移植到正常微环境后得以修复,从而证明了BM基质的积极作用。与患者骨质疏松症的常见发现一致,研究人员发现,随着甲状旁腺素(PTH)水平的降低,骨沉积减少;甲状旁腺素(PTH)不仅是骨代谢的关键调节因子,还是HSC活性的重要调节因子。

在体内,通过重建的Jagged1和骨桥蛋白水平激活PTH信号与通过纠正HSC-壁龛交联挽救th3 HSCs的功能池相关。在地中海贫血患者中证实了静息态的HSC减少,且伴随BM基质壁龛的特性改变。

本研究揭示了β-地中海贫血患者由BM微环境改变引发的HSCs缺陷,为改善移植和基因疗法提供了新的见解。
