免疫检查点抑制剂是一类能阻断某些蛋白质的药物,阻断蛋白质后会松开某些免疫细胞的“刹车”,使它们能够攻击癌细胞.。


研究发现,ANGPT2也可能成为免疫检查点和血管生成抑制剂的组合疗法的靶点。


“在这项研究中,我们发现ANGPT2是机体对免疫检查点CTLA-4和PD-1抑制剂反应的预测因子和预后标志物,”说达纳-法伯癌症研究所黑色素瘤中心的主任F. Stephen Hodi说:“我们发现了更多的证据。这些证据表明,血管生成因子在免疫调节、靶向免疫系统中的作用以及血管生成因子与癌症疗法进行组合的可能性。”


在这项研究中,Hodi和他的同事们研究了血管生成因子ANGPT2在免疫调节中的作用,及将其作为免疫检查点抑制剂的生物标志物的潜力。ANGPT2血管生成的调节剂,并使血管成熟。在之前的研究中,该蛋白与抗贝伐单抗(Avastin)抵抗有关。抗贝伐单抗(Avastin)是靶向针对另一种血管生长因子(VEGF)的治疗性抗体。


为了理解这ANGPT2影响临床预后的机制,研究人员进行了在实验室基础上进行了测试,使用巨噬细胞(一种免疫细胞)特定的亚群,发现ANGPT2能增加这些细胞内PD-L1的表达。这种亚群的巨噬细胞在肿瘤微环境中有被发现。PD-L1是一种蛋白质,在免疫抑制中发挥着作用,且有一些免疫检查点抑制剂以PD-1/PD-L1轴为靶点。


这些体外研究表明,血管生成因子不仅在血管形成中起作用,还有免疫调节效应。因此,以血管形成为靶点的疗法或可与免疫检查点封锁发挥协同作用。目前还需要进行更大规模的试验来进一步验证这些结论。
