研究显示,内分泌干扰物(EDC)会产生生殖功能障碍,影响类固醇激素合成和类固醇合成相关基因的表达,然而相关研究主要是关于暴露于单一化合物对人类健康的影响。而实际上,人类暴露于EDCs混合物的环境中。因此研究人员评估了慢性暴露于单一烷基酚和邻苯二甲酸酯以及他们的混合物对小鼠睾丸组织和类固醇生成的影响。

妊娠小鼠通过饮用水暴露于:单一邻苯二甲酸酯(DEHP,DBP,BBP)0.3mg/Kg体重(BW)/天;单一烷基酚(NP,OP)0.05mg/Kg体重(BW)/天,以及他们的混合物。暴露时间为从交配后0.5天到仔鼠断奶,雄性后代继续暴露直至成年(60天龄)。

结果显示,暴露于混合物的小鼠身体和相对睾丸重量增加,伴随组织学改变。睾丸内睾丸素(T)仅在接触DBP的小鼠中发生变化,而雌二醇(E2)水平在所有组中均被改变(BBP除外)。暴露于单一或混合化合物的小鼠编码类固醇途径激素基因(Cyp11a1,Hsd3b1,Cyp17a1和Cyp19a1),胆固醇转运蛋白基因(Star)和转录因子(Sp1)基因的mRNA水平中至少有两个发生改变,然而不同类型的暴露并不是会引起所有相关基因表达的变化。并且单一化合物在mRNA或蛋白质水平上的变化并不总是与混合物相同。

该研究表明,慢性暴露于EDCs混合物对类固醇合成途径和激素相关的基因和蛋白质表达的影响不同于暴露于单一EDC,这对于以后的相关研究提供重要参考。
