Neurology:成人散发性-退行性共济失调的临床和遗传学特征

成人散发性共济失调的临床表型和进展进行了定量表述,并对6%的患者致病突变进行了确证

近日研究人员对成人散发并发退行性共济失调的临床特征、病程以及致病突变因素进行了研究。

研究采用共济失调的评定标准(SARA)衡量患者疾病严重程度。 采集患者DNA样本,采用高覆盖性共济失调特异基因板以及下一代测序技术进行基因突变筛查

研究包含了249名患者,其中83人临床符合多系统小脑萎缩型(MSA-C)共济失调。MSA-C阳性以及病程与SARA得分相关。48名患者不完全符合MSA-C 指征且病程超过10年,该类患者被定义为不明原因成人散发性非多系统小脑萎缩型共济失调(SAOA/non-MSA)。与MSA-C患者相比,该类患者SARA分数更低 (13.6 ± 6.0 vs 16.0 ± 5.8, p = 0.0200) 且每年的SARA分数增加较慢(1.1 ± 2.3 vs 3.3 ± 3.2, p = 0.0013)。所有患者中,6%的患者存在明确或可能的基因诊断

研究首次对成人散发性共济失调的临床表型和进展进行了定量表述,并对6%的患者致病突变进行了确证。
