Am J Resp Crit Care:早产儿血管内皮细胞线粒体功能预测死亡或肺部转归!

血管内皮细胞线粒体功能障碍参与了多种氧化应激相关疾病的发病机制。氧化应激是支气管肺发育不良(BPD)发病机制的一个主要因素,BPD是一种慢性的早产儿肺疾病,常导致成年幸存者后遗症。近日,呼吸内科以及重症医学领域权威杂志Am J Resp Crit Care上发表了一篇研究文章,研究人员确定从极度早产的婴儿获得人脐静脉内皮细胞中线粒体生物产能功能和氧化剂产生的差异是否与BPD和末次月经后36周内死亡风险相关。

研究人员测定了69例婴幼儿脐静脉内皮细胞耗氧量、超氧阴离子和过氧化氢的产生。

相比于从无BPD的婴儿获取的人脐静脉内皮细胞,从BPD或死亡婴儿获取的人脐静脉内皮细胞有较低的最大OCR(平均±SEM为107±8 vs. 235±22皮摩尔/分钟/ 30000细胞,P<0.001),高氧暴露后产生更多的超氧阴离子(平均±SEM为:89807±16616 vs. 162706±25321 MitoSOX红色荧光单元,P<0.05)并且高氧暴露后释放更多的过氧化氢(H2O2)到上清液中(平均±SEM为1879±278 vs. 842±119荧光的单位,P<0.001)。

该研究结果表明,迟发型BPD或死亡的早产儿的内皮细胞中线粒体生物产能受损,并且在出生时产生更多的氧化剂,表明这些婴儿出生时的血管内皮细胞线粒体功能障碍一直持续到他们产后的生命,并有助于肺不良结局产生和早期死亡率的增加。
