为了转移,癌细胞必须首先成为活动性的,然后爬过周围组织抵达血管。在新的研究中,MIT研究团队发现,癌细胞会跟随纤连蛋白的足迹。纤连蛋白是"细胞外基质"的一部分,为周围细胞提供支持。纤连蛋白在肿瘤边缘和附近血管位置的浓度特别高。

"肿瘤环境中的癌细胞不断面对纤连蛋白浓度的差异,他们需要能够从低浓度移动到高浓度以抵达血管"Oudin说道。

MenaINV是正常Mena蛋白质的一种替代形式,在该过程中扮演关键角色。MenaINV包含一个正常版本中没有的节段,这使其更强烈地结合到受体alpha-5整合素。alpha-5整合素存在于肿瘤细胞和附近支持细胞的表面,并识别纤连蛋白。

当MenaINV附着到这种受体,它会促进纤连蛋白结合到相同的受体。纤连蛋白通常是一种缠结蛋白质,但当它结合到细胞表面,则会伸展成为长束。这会刺激胶原组织(另一种细胞外基质蛋白)成为僵硬纤维,从肿瘤边缘辐射出来。

这种在更恶性肿瘤中通常可见的模式,基本上为肿瘤细胞移动到血管铺平了道路。

"如果是卷曲、盘绕的胶原蛋白,那么会是好的结果。但是,如果被重新排列成这些直的长纤维,则会为细胞迁移提供高速公路"Oudin说道。

在小鼠研究中,携带侵润形式Mena的细胞能够更好地识别和爬向更高浓度的纤连蛋白,沿着胶原蛋白通路移动,而没有MenaINV的细胞则没有向更高浓度移动。

研究人员还查看了乳腺癌患者的数据,发现高水平的MenaINV和纤连蛋白与转移和早期死亡相关。然而,正常版本的Mena和早期死亡之间没有联系。

Gertler的实验室之前开发出能够检测正常和侵润形式Mena的抗体,现在正在开发用于测试患者活检样本。这些测试可以帮助医生确定患者肿瘤是否可能会扩散,甚至可能指导对患者的治疗。此外,科学家们也许能够开发出抑制MenaINV的药物,这可能有助于治疗癌症或防止其转移。

研究人员们现在希望研究MenaINV如何可能促进其他类型的癌症。初步研究表明,它在肺癌和结肠癌中扮演着类似于乳腺癌中的角色。他们也将研究如何调节两种形式的Mena蛋白质,以及细胞外基质中的其他蛋白质如何可能促使癌细胞迁移。
