JCLA:新的检测方法可保持原比例和完整的尿细胞游离DNA?

尿游离DNA (ucfDNA)由于核酸酶活性高,且尿液中含有复杂物质,因此容易发生降解。因此,我们开发了标准化的尿液收集管(UCT),以防止ucfDNA的降解,同时在收集样本的过程中保持尿液细胞的原始形态,确保了ucfDNA原始比例的稳定和完整性。 研究人员分别分用10‐mL正常管和10‐mL UCTs收集膀胱癌患者的尿液样本,并置于室温下保存。离心分离尿上清,提取ucfDNA。然后用定量实

尿游离DNA (ucfDNA)由于核酸酶活性高,且尿液中含有复杂物质,因此容易发生降解。因此,我们开发了标准化的尿液收集管(UCT),以防止ucfDNA的降解,同时在收集样本的过程中保持尿液细胞的原始形态,确保了ucfDNA原始比例的稳定和完整性。

研究人员分别分用10mL正常管和10mL UCTs收集膀胱癌患者的尿液样本,并置于室温下保存。离心分离尿上清,提取ucfDNA。然后用定量实时聚合酶链反应对ucfDNA进行定量。用Agilent 2200分析UcfDNA片段的分布,用nextgeneration测序方法检测泌尿系统疾病特异性突变的频率。

UCTs中收集到尿液在常温下,在7天内ucfDNA原比例和完整性无统计学显著变化,且ucfDNA片段保存良好。相反,收集到正常尿管中的尿液,其原始ucfDNA比例明显下降,ucfDNA片段变化较大。UCTs基因特异性ucfDNA等位基因的△% (AF)比正常试管低3.7倍。

研究表明,利用UCTs技术,可以最大限度地保持ucfDNA的原始比例和完整性,稳定尿细胞,并将尿细胞DNA释放引起的背景噪音降到最低,这将有助于利用下一代利用尿液进行无创液体活检的应用。
