研究人员采用亚克隆选择计算模型和适用于高通量测序数据的理论种群遗传学来监测人类癌症克隆动态,这种方法可以察觉肿瘤样本的亚克隆结构,同时测量每个亚克隆的选择性优势和出现时间。

“知道肿瘤是否会以某种方式改变,据此设计有效治疗,”伦敦玛丽王后大学Barts癌症研究所教授Trevor Graham说。“这项工作还处于起步阶段,待其成熟后将变得非常强大。”

解读肿瘤的“秘密日记”

随着肿瘤生长,它们的遗传密码(突变)也在不断改变。其中一些变化可能赋予癌细胞优势,使其更适合在某环境下生存。遵循达尔文进化论原则,这样的细胞受到青睐能够保持复制,产生更多相同突变细胞。许多携带不同突变的细胞群都在如此生长和聚集,创造出具有高度遗传多样性的肿瘤。

我们知道癌症富于变化,却无法得知都发生过哪些确切变化。因为所有被送到实验室分析的肿瘤样本都属于能在患者体内被检测到的样本。因此,我们看不见真实肿瘤的生长历史,这项研究则致力于填补这些缺失信息。

通过解读肿瘤细胞的基因序列,人们尝试用计算机系统读取隐藏在基因组突变中的“秘密日记”。值得注意的是,这项研究揭示细胞获得的突变驱动癌症进程,这种突变被命名为驱动突变(driver mutations),它们让细胞在某些情况下比其他细胞的增长速度快30%。

用肿瘤的历史预测它的未来

计算机系统利用这些遗传数据和定义算法预测癌症的未来进化变异。它将进一步推进“智能诊断”,并为个性化医疗提供巨大助力。

在这项研究转化为临床实践之前,研究团队首先需要验证系统预测的正确性。他们应用来自乳腺、胃、血液、结肠、和肺等癌症样本和转移性沉积物的高深度测序数据,表明选择进化而来的可检测亚克隆在肿瘤生长过程早期始终如一的出现,而且具有较大的适应优势(>20%)。此后,他们还将运用系统进行临床前模型和长期监视下的患者肿瘤样本分析。

论文第一作者Marc Williams 补充道:“我们想要测量癌症进程变化,这些变化往往很难量化。通过方方面面的测量和分析,我们的远期目标是实现对癌症发展轨迹给出重要预测。”
