J Orthop Surg Res:评估掺入羟基磷灰石的聚醚醚酮生物复合材料的生物活性

在该研究中,使用复合和注塑技术将羟基磷灰石(HA)掺入PEEK中以制造HA/PEEK生物复合材料。测试制备的HA/PEEK复合材料的拉伸性能(HA含量为0-40重量%)以选择最佳HA含量。为了评价复合物的生物活性,评估MC3T3-E1细胞的细胞附着、增殖、扩散和碱性磷酸酶(ALP)活性,以及浸入模拟体液(SBF)后的磷灰石形成,以及兔颅骨缺损模型中的骨整合。将结果与超高分子量聚乙烯(UHMWPE)和纯PEEK的结果进行比较。

结果显示,通过评估具有不同HA含量的PEEK复合材料样品/ PEEK复合材料的拉伸性能和弹性模量,选择30wt%HA / PEEK复合材料用于随后的测试。细胞试验结果表明,与UHMWPE和纯PEEK相比,PEEK复合材料样品/ PEEK复合材料具有更好的细胞附着、增殖、扩散和更高的ALP活性。在SBF中浸泡7天后,在HA/PEEK复合材料上形成磷灰石岛,并且在较长时间内连续生长。动物试验表明,HA/PEEK复合材料周围的骨接触和新骨形成比UHMWPE和纯PEEK周围的骨接触和新骨形成更明显。

综上所述,该研究结果表明,通过复合和注射成型技术产生的HA/PEEK生物复合材料表现出增强的成骨作用并且可以用作整形外科植入物的候选者。
