英国伯明翰大学的研究人员发表在Chemical Science上的一项研究结果表明,新技术可减少传统诊断方法的假阳性问题。

研究者 Prof. Paula Mendes表示,新型传感器具有两大关键优势:

“对患者来说最重要的是,这种传感器可提供更准确的读数并可减少假阳性结果。此外,我们的技术也仅是简单的生产和存储,所以可以广泛存在全世界医生的手术架上。传感器还可回收多向应用,且依旧可保持其准确性。”

据美国癌症协会(ACS)报道,前列腺癌是造成美国男性死亡的第二大癌症,38名癌症死亡患者中就有1名死于前列腺癌。据估计,2015年美国将有220800名新发前列腺癌患者。

前列腺癌通常依赖于抗体的检测,但是这种检测的费用极高且易受环境的影响。最重要的是,假阳性率高。

研究人员认为,他们的新型传感器芯片可以提高前列腺癌的诊断,避免这些不利影响。

由化学工程师及化学家所创造的传感器芯片可识别称为糖蛋白分子的特异性结合糖链。这些分子在机体中发挥多种功能,包括免疫反应。

由于糖蛋白分子在免疫反应中的作用,科学家经常使用其作为疾病(包括前列腺癌)的生物标志物。以往用于检测糖蛋白的技术主要集中在分子的蛋白部分,然而,这些分子在身体受到疾病影响时并不总会发生改变。

相反的是,这种新型传感器芯片设计的重点在于分子的碳水化合物部分,而这些碳水化合物可在健康和患病个体之间略有不同。

芯片技术还可用于其他疾病的诊断

“它们并不依赖于患者的自觉症状,故生物标志物(如糖蛋白)的检测必不可少,这是检测的结果模棱两可,而且生物标志物可能不会再疾病刚发生时便可出现,”Prof. Mendes解释道,“但是,生物标志物的变化却极小,特异性也不够,所以我们需要可以分辨出这些抗体所不能是别的细微差别的技术。”

新型传感器芯片表明存在可与特异性前列腺癌相关蛋白结合的纳米孔。这些孔是由专门设计的可结合在金属表面及前列腺癌蛋白的分子构成。分子末端包含可与糖蛋白反应的硼族,当这种硼结合被打破时,一个完美的小孔便出现了。

Prof. Mendes继续解释道:

“纳米孔本身像一把锁,关键是找到特定的前列腺蛋白。可能存在合适大小的糖蛋白分子,但是它们不能与特异性组合的硼族结合。”

该研究小组希望可与商业伙伴进行进一步的研究与合作,这可使得他们的新技术应用于所有前列腺癌的检查,并可适用于其他的治疗条件。

“我们相信,这可应用于其他诊断所存在的挑战,”共同作者Dr. John Fossey说道。“很多疾病会产生特异性的糖蛋白,因此存在一种方法可提高我们诊断的准确性。”


与其他任何癌症一样,可靠且快速的诊断对确保患者获得最佳治疗的来说至关重要。这项新技术不仅在前列腺癌诊断上而且也在其他疾病的诊断上具有非法的成就。
