Sci Rep:人类膀胱癌细胞中原苏木素B能够促进细胞凋亡和引起G1细胞周期阻滞

最近,有研究人员调查了原苏木素B对膀胱癌细胞的细胞增殖和细胞凋亡的影响。研究人员利用MTT试验对原苏木素B的不同浓度(12.5, 25, 50, 100或者200μg/mL, 48h)对SV-HUC-1, T24和5637细胞增殖的影响进行了评估,并利用流式细胞术对原苏木素B的不同浓度(100, 150, 200, 250和300μg/mL, 48h)对细胞凋亡和细胞周期的影响进行了分析。研究发现

最近,有研究人员调查了原苏木素B对膀胱癌细胞的细胞增殖和细胞凋亡的影响。研究人员利用MTT试验对原苏木素B的不同浓度(12.5, 25, 50, 100或者200μg/mL, 48h)对SV-HUC-1, T24和5637细胞增殖的影响进行了评估,并利用流式细胞术对原苏木素B的不同浓度(100, 150, 200, 250和300μg/mL, 48h)对细胞凋亡和细胞周期的影响进行了分析。

研究发现,200μg/mL原苏木素B处理T24和5637细胞表现出了形态上的变化(收缩、变圆、膜异常和粘连减少),但是原苏木素B对SV-HUC-1细胞的增殖停滞没有影响。另外,原苏木素B能够引起浓度依赖的细胞生长抑制,在T24细胞中IC50为82.78μg/mL,在5637细胞中为113.79μg/mL。原苏木素B还能够引起T24和5637细胞系浓度依赖的细胞凋亡的增加(100-300μg/mL)。原苏木素B在上述2种细胞系中对细胞周期中的影响均为G1期停滞,并且S期细胞的比例和增殖指数减少。蛋白组分析表明了原苏木素B能够调控许多参与细胞周期的基因。

最后,研究人员指出,原苏木素B能够以浓度依赖的方式抑制人类膀胱癌T24和5637细胞系中的增殖和促进细胞凋亡,很可能是通过干扰细胞周期调控从而阻止G1到S期转换来实现。
