库欣病（Cushing Disease）是由大脑中脑垂体肿瘤引起的。肿瘤过多分泌应激激素促肾上腺皮质激素（ACTH），随后肾上腺释放皮质醇，导致体重迅速增加，血压升高和肌肉无力。库欣病患者容易发生骨质疏松症，感染和认知功能障碍，甚至抑郁症。在80至85％的库欣病患者中，脑垂体肿瘤可以通过脑外科手术去除。

对于不能手术的情况，目前只有唯一一个有针对性的治疗，但不幸是会引起剧烈的副作用， 20％以上患者会有高血糖。研究领导者内分泌学家Günter Stalla利用细胞培养，动物模型和人类肿瘤组织，发现一个无害的植物提取物可用于治疗Cushing病。
 
水飞蓟宾是水飞蓟种子的主要活性成分，对于人类它具有卓越的安全性，并已用于治疗肝脏疾病和中毒。新研究结果发表在Nature Medicine杂志。经过水飞蓟宾处理，肿瘤细胞的ACTH生成恢复了正常，肿瘤生长速度减慢，老鼠库欣病症状消失。
 
在100000人中仅有5.5人会罹患库欣病，但这个疾病在宠物中是很常见的。例如，4％的狗，甚至7％的马匹患有库欣病。
 
我们知道，库欣病是由过多的促肾上腺皮质激素释放引起的。但是什么原因导致促肾上腺皮质激素过量生成以及如何阻止它却不得而知。在实验中，研究人员发现，热休克蛋白90（HSP90）在库欣病患者肿瘤组织中具有非常高的量。正常量HSP90有助于正确折叠另一种蛋白质--糖皮质激素受体，继而抑制生产促肾上腺皮质激素。
 
由于有太多的HSP90分子在肿瘤组织中，它们与糖皮质激素受体结果，Paez-Pereda解释：我们发现，水飞蓟宾能结合HSP90，从而将糖皮质激素受体分子从HSP90上“释放”出来。水飞蓟宾或许可以提供一种非侵入性治疗策略，不仅对库欣病，甚至对糖皮质激素受体参与的其他疾病如的肺肿瘤，急性淋巴细胞性白血病或多发性骨髓瘤等也有益处。
