科学家们最近发现一个参与DNA包装的基因开关可能是癌细胞获得永生能力的关键。相关研究结果发表在国际学术期刊Science上。弗朗西斯克里克研究所的研究人员在Cancer Research UK的部分资助下发现一种叫做H1.0的蛋白表达在许多癌症类型中经常发生关闭,重新激活这种蛋白的表达能够阻止肿瘤的生长。通过对缺少H1.0的癌细胞进行研究,他们发现DNA在一些关键位置处于展开状态,导致一系列基因发


科学家们最近发现一个参与DNA包装的基因开关可能是癌细胞获得永生能力的关键。相关研究结果发表在国际学术期刊Science上。

弗朗西斯克里克研究所的研究人员在Cancer Research UK的部分资助下发现一种叫做H1.0的蛋白表达在许多癌症类型中经常发生关闭,重新激活这种蛋白的表达能够阻止肿瘤的生长。

通过对缺少H1.0的癌细胞进行研究,他们发现DNA在一些关键位置处于展开状态,导致一系列基因发生激活使癌细胞处于“永生”状态。癌细胞因此持续分裂,促进了肿瘤的生长。

但是随着肿瘤的生长,一些细胞内的H1.0会自发地重新开启表达。研究人员对这种现象进行追踪,找到一个能够控制H1.0蛋白合成开启的DNA区域。随着H1.0重新表达发挥作用,细胞需要用来保持分裂的基因会重新被关闭,使细胞重新回到正常的有限寿命中。

研究人员现在正在寻找能够使所有癌细胞都重新开启H1.0蛋白合成的药物,加速上述过程。这将为阻止多种类型肿瘤的生长提供一种有效方式。

领导该项研究的Dr Paola Scaffidi这样说道:“这项研究提出了通过将癌细胞逆转回有限寿命状态,最终引起肿瘤停止生长,将恶性肿瘤重新变成良性瘤的可能。我们还发现肿瘤中H1.0表达水平很低的病人其病情趋于更加恶化,并且在多种类型的肿瘤病人中都是如此。”

“我们现在知道从哪里开始寻找通过消除癌细胞的永生能力发挥作用的药物,而不仅仅是杀死他们。”

另一位研究人员表示,癌细胞通过改变DNA包装干扰一些基因的活性,靶向这条途径的药物是一个令人兴奋的新研究方向,一些治疗药物已经进入临床用于白血病治疗。这个研究首次揭示了这种方法如何在实体瘤中发挥作用,也让我们对组成肿瘤的不同细胞类型有了更深一层的理解。

晁明:喝碱性水有益于健康是公认的,但对于“苏打水防癌作用”这个问题,目前还没有准确的科学依据。

我们采用的方式是把碳酸氢钠通过肿瘤滋养血管注入瘤体,改变肿瘤内部的pH值。用碱如碳酸氢钠(小苏打)来去除肿瘤内的氢离子,就可破坏乳酸根和氢离子的协同作用,从而快速有效地杀死处于葡萄糖饥饿或缺乏的肿瘤细胞。
