Circulation.:长非编码RNA MANTIS促进内皮血管生成功能

内皮细胞的血管生成功能受到许多机制的调节,但是长非编码RNA(lncRNA)对其的影响尚未被研究。近期德国学者发表研究文章于著名血管杂志Circulation,以发现新型的功能重要的内皮细胞lncRNA。

通过表观遗传学控制的内皮lncRNAs检索发现lncRNA n342419,这里称为MANTIS,作为最强的调节lncRNA。由组蛋白去甲基化酶JARID1B控制,MANTIS在特发性肺动脉高压患者和野百合碱处理的大鼠中表达下调,但是在动脉粥样硬化饮食的猕猴属颈动脉以及从人胶质母细胞瘤患者中分离的内皮细胞中表达上调。使用siRNA或GapmeR的CRISPR / Cas9介导MANTIS的缺失或沉默可抑制剪应力损伤时的内皮细胞血管萌出和排列。机制方面,核定位发现MANTIS lncRNA与SWG / SNF染色质重塑复合物的催化亚基BRG1相互作用。这种相互作用是通过保持BRG1的ATP酶功能活性的核小体重塑所必需的。因此,关键性内皮基因SOX18,SMAD6和COUP-TFII等的转录是通过有效的RNA聚合酶II的机械性结合来调节的。MANTIS是一种差异性调控的新型lncRNA,具有促进内皮血管生成的功能。
