MJ:一种用于减少恶性疟原虫抗体多重测定中板间差异的分析方法

抗体在疟疾免疫中发挥重要作用。最近的研究表明,针对多种抗原的抗体以及总体反的应范围均与疟疾的防治有关。然而,使用多重测定法对疟疾抗体组合测量的变异性和可靠性尚未得到很好的验证。本项研究使用了混合阳性和阴性对照重复来减少板间差异的一种标准化程序。

抗体在疟疾免疫中发挥重要作用。最近的研究表明,针对多种抗原的抗体以及总体反的应范围均与疟疾的防治有关。然而,使用多重测定法对疟疾抗体组合测量的变异性和可靠性尚未得到很好的验证。本项研究使用了混合阳性和阴性对照重复来减少板间差异的一种标准化程序。研究通过9种疟疾抗原,包括裂殖子抗原(AMA1,EBA175,MSP1 ,MSP2,MSP3,MSP11,Pf41),子孢子CSP和怀孕相关的VAR2CSA的抗体水平的检测,对60个测试样品(30个来自疟疾阳性个体和30个疟疾阴性个体)以及5个混合阳性对照和2个混合阴性对照进行筛查。在3个板中以一式三份测量抗体水平,并且通过相同的技术人员在两个不同日期进行重复实验。通过在线性和自然对数尺度对混合样本的测试来评估所提出的标准化程序的性能。与线性尺度上的数据相比,自然对数变换数据偏移较少并且平均方差关系减弱。对混合对照采用自然对数尺度的标准化程序能够显著降低板间差异。对于通过多重测定法测量多抗原抗体的疟疾相关研究,研究推荐使用自然对数转换进行数据分析和具有多个混合样本的标准化程序可以提高抗体检测的精度。 原始出处: Fang R,et al.An a
