J Hepatology: 基因组测序确定WNK2是肝细胞癌的驱动因素和早期复发的危险因素

背景与目标

方法
研究人员对182例原发性肝癌样本进行了全基因组测序(49例)以及深度靶向测序(115例),特别关注WNK2基因位点,使用Sanger测序和qPCR评估了另外554个HCC样品中该基因的所有编码外显子和拷贝数。还探讨了WNK2对肿瘤生长和转移的功能作用和机制。

结果
研究人员在182例原发性HCC样本中鉴定了5个基因位点(WNK2,RUNX1T1,CTNNB1,TSC1和TP53),它们具有与治愈后早期肿瘤复发相关的体细胞突变。在专注于WNK2的736例HCC样本中,总的体细胞突变和拷贝数损失分别发生5.3%(39/736)和27.2%(200/736)。两种类型的变异都与较低的WNK2蛋白水平,较高的早期肿瘤复发率和较短的总生存期有关。生物功能研究揭示了WNK2的肿瘤抑制作用:其失活导致HCC细胞中ERK1 / 2信号激活,肿瘤相关的巨噬细胞浸润以及肿瘤生长和转移。

结论
本项研究证明WNK2基因为根治性切除术后早期HCC复发的驱动因素。
