J Endod:生物陶瓷材料对人根尖牙乳头干细胞的增殖和分化影响

在牙髓再生治疗(RET)过程中,医师偏爱使用生物陶瓷作为置于血块上方的封闭材料。然而,清楚和理解根管内封闭材料与细胞之间的相互作用却十分重要。这篇研究的目的是为了比较不同生物陶瓷材料(ProRoot MTA [Dentsply, Tulsa, OK], Biodentine [Septodont, Saint-Maur-des-Fossés, France], and RetroMTA [BioMTA, Seoul, Korea])做为封闭材料在RET中对人根尖牙乳头干细胞(SCAPs)增殖和分化的影响。

研究将SCAPs按照20,000 cells/well接种于培养皿中,通过transwell培养皿放置水溶性的测试材料以此对细胞施加刺激。采用3-(4,5-二甲基噻唑-2-基)-2,5-二苯基四唑溴化物测定在1, 3, 7和14天时检测SCAP的增殖情况。通过茜素红染色和实时定量聚合酶链式反应在不同时间节点(1, 7, 14和21 days)检测SCAP的分化情况。研究所检测的成牙本质细胞基因的表达包括:牙本质基质酸性磷蛋白1、牙本质涎磷蛋白、骨钙素和基质细胞外磷酸糖蛋白。另外,SCAP在成牙本质/成骨诱导培养基下培养时也施加了水溶性测试材料刺激。

结果显示,所有3种测试材料均可诱导SCAP增殖。与对照组相比,Biodentine, ProRootMTA和RetroMTA组在培养第7和14天均显著诱导SCAP的增殖。在成牙本质细胞分化检测中,仅有Biodentine展现出茜素红染色阳性。在培养21天的Biodentine组中可检测到牙本质基质酸性磷蛋白1、牙本质涎磷蛋白和基质细胞外磷酸糖蛋白的最高表达。7天时,可检测到骨钙素表达显著上调。

结论:Biodentine, ProRootMTA和RetroMTA可以诱导SCAP的增殖。在3种材料中,Biodentine可以明显诱导SCAP的分化。
