Nature:组织蛋白酶B介导辐射诱导的旁观者效应

辐射诱导的旁观者效应(RIBE)特指被辐射的细胞或组织释放的因子对未暴露于辐射中的动物的其他部分产生影响。辐射诱导旁观者效应会引起基因组的不稳定性,应激反应,以及细胞凋亡或细胞增殖的改变。

虽然辐射诱导的旁观者效应对于放射防护,辐射安全和放射治疗具有重要意义,但是至今为止,关于参与其中的因子,以及其作用机制的了解依旧很少。

来自清华大学课题组的研究人员利用秀丽隐杆线虫作为研究辐射诱导旁观者效应的模型,发现了第一个线虫中参与辐射诱导旁观者效应的因子CPR-4。CPR-4是人类组织蛋白酶B的同源物。CPR-4是紫外线或电离γ射线照射的动物分泌的,也是条件培养基中抑制细胞死亡或是增加未照射动物胚胎致死率的主要成分。研究人员发现,CPR-4导致在照射组织远端的未暴露部位出现类似的现象,以及应激反应的增加。辐射之后, CPR-4的活性由p53同源物CEP-1调节,CPR-4似乎通过作用于胰岛素样生长因子受体DAF-2发挥辐射诱导的旁观者效应。

我们的研究为辐射诱导的旁观者效应提供了重要的理解,并有助于确定其他辐射诱导的旁观者效应因子及其作用机制。
