Nature:陈志坚团队:解开先天免疫领域的一大“谜团”

11月29日,前不久刚获得“生命科学突破奖”的陈志坚教授又有好消息,其团队在最新一期《Nature》发表了重要研究成果,解开了先天免疫领域一个存在已久的谜团。

一直以来,科学家们试图弄清楚一种炎症小体——NLRP3是如何在多种刺激下促进炎症反应的。以往研究揭示,NLRP3炎症小体在病原体感染、阿尔兹海默症、癌症、2型糖尿病等多种疾病中都发挥着举足轻重的作用。遗憾的是,这背后的机制一直未能破解。

现在,来自德克萨斯大学西南医学中心的陈志坚研究员带领团队解开其中的关键。他们通过生化、成像和遗传学方法解析了NLRP3炎症小体的结构变化,发现不同的刺激会导致细胞内的高尔基体反面网络结构(trans-Golgi network,TGN)发生解体,形成巨大的囊泡。

这些囊泡中含有一种特殊的脂质成分(PI4P),会与NLRP3的特定区域结合,进而激活NLRP3,启动相应的炎症反应。

“NLRP3炎症小体的独特之处在于,它可以被大量刺激触发。我们的研究发现,NLRP3并不会直接识别有毒物质,而是通过检测细胞结构变化(由引发细胞损伤的不同刺激造成)完成响应。” 陈志坚解释道。
