Sci Rep:人类前列腺正常细胞和癌细胞外代谢组学区分

血清前列腺特异性抗原(PSA)是目前最常用于前列腺癌(PCa)检测临床实践的生物标记。然而,该生物标记具有一些缺陷。最近,有研究人员基于无靶标气相色谱-质谱分析法(GC-MS)对PCa细胞的代谢组进行了分析,从而来验证在PCa细胞系中,与正常前列腺细胞相比,代谢情况发生变化。

研究人员利用GC-MS技术,评估了4个前列腺细胞系和1个正常前列腺细胞系细胞外基质(外代谢组)的挥发性有机化合物差异(VOCs),并且是在2个pH值下进行的(pH值分别为2和7)。多变量分析表明,一组挥发性代谢产物能够区分癌化细胞和正常前列腺细胞。其中变化最多的代谢产物包括酮、醛和有机酸。研究人员重点强调了正十五烷酮和葵酸,这两种物质在PCa细胞中与正常细胞相比,显著增加;并且环己酮、6-甲基酮、2-甲基戊烷-1,3二醇、4-甲基苯甲醛、苯甲酸甲酯、壬酸等有机物在PCa细胞中显著减少。另外,尽管区分能力相似,PCa挥发性物质能够被VOCs提取pH显著影响。

最后,研究人员指出,他们的数据表明了VOCs具有作为PCa筛选方法学上潜在物质。
